Biopharma companies use various techniques to generate, isolate, and characterize antibodies with desirable functionality and specificity. The most promising antibodies are then selected for further study and undergo cloning and expression to produce proteins for downstream development and testing.
The Sapio Sciences antibody workflow solution allows scientists to capture the workflow from start to finish—from immunization of the animals with the target antigen through sample collection, antibody isolation and characterization, cloning and expression of selected candidates, and protein production and purification. Each workflow step is captured in one or more templated experiments, which are then executed sequentially by the research teams involved.
The workflows are flexible to accommodate a variety of methodologies. For example, antibodies can be isolated and characterized using various techniques, including hybridoma, phage display, or B-cell sorting. Each has its advantages and disadvantages around specificity, scope of targets, and throughput. Each also has its workflow, which the solution accommodates. For example, when using a hybridoma approach, it is necessary to perform primary and secondary in-vitro screening to filter for the most promising candidates. On the other hand, B-cell sorting using flow cytometry and microfluidic approaches enables the direct isolation of the most promising candidates.
The solution fully integrates with the Sapio biologics registration and materials management system. At every step of the workflow, entities are created and consumed, automatically registered, and tracked in the entity registries. At the end of the process, an entire lineage of any entity and its samples can be traced back to its initial identification and/or production.

Flexible Workflow
The out-of-the-box antibody workflow consists of several steps, each with its own experiment template. The workflows and the template at each step are easy to modify using Sapio’s no-code configuration capabilities. This allows you to accommodate variations in your process, such as using hybridoma vs. B-cell sorting in the initial antibody discovery phase.
A Scientific Ecosystem
The antibody workflow is just one element of the Sapio Sciences fully integrated collection of scientific solutions. For example, Antibodies produced in this workflow may then be used to produce Antibody Drug Conjugates in another workflow, combining with linkers and payloads produced in a medicinal chemistrty workflow. Downstream, the final antibodies may be sent to a bioprocessing workflow as they move into development and manufacturing
Registration and Lineage
The workflow is built on top of the biologics registration system. This allows every entity (antibody, plasmid, tissue, etc) that is created in the workflow to be registered directly from the workflow experiments. Every sample of those entities created or consumed is also captured and tracked automatically. At any stage in the process, you can see the entire lineage of an antibody or any other entity laid out in a clear graphical form, providing complete traceability
Enterprise-Wide Collaboration
Many teams are involved in discovering and developing an antibody, including molecular biologists, cell biologics researchers, immunologists, protein scientists, structural biologists, etc. These teams may be in-house or at external research partners such as CROs. The Sapio workflow solution allows requests, experiments, and sample information to flow seamlessly between one team and the next as the project progresses. Project leaders can monitor progress and assign work using graphical dashboards.
